Adam Copley and Liam Johnson took three wickets for Barnsley.
The Shaw Laners are one place and 17 points clear of the relegation zone with five games left. Cawthorne - who have done the double over Barnsley in the league as well a cup win - are four places clear of the drop zone, and 29 points.
Cawthorne then lost by a wicket at home to Castleford in the K3 Dental Cup.
Defending 154, Danish Aziz took three wickets but the visitors snuck into the final.
Bottom club Elsecar suffered an eighth straight league loss.
They were skittled for 108 chasing 298 at Appleby Frodingham.
Elsecar are 44 points from safety so could be relegated over the Bank Holiday weekend.
